Accelerating the Proteomics Discovery to Quantification Pipeline

The need to identify and accurately quantify a protein or a panel of proteins in multiple samples is becoming increasingly important in the life sciences, pharmaceutical, clinical and food and beverage industries. But which protein to analyze may need a continuous research pipeline where key proteins in a biological system are discovered, and then verified and validated in greater numbers of samples to account for biological variation. Ultimately assays are developed for routine detection of the protein.
Discovery Proteomics using ‘shotgun’ mass spectrometry approaches generate many protein candidates but do not generally have the ability to quantitatively analyze large numbers of samples. Multiple reaction monitoring (MRM) mass spectrometry assays are relatively cheap and fast to develop, and the quantitative assays are reproducible and accurate. They can be performed in high throughput, and many proteins can be assayed in the same experiment.

Dr. Amanda Paulovich earned her B.S. in Biology from Carnegie Mellon University in Pittsburgh, and her Ph.D. and M.D. degrees from the University of Washington in Seattle. She completed training in Internal Medicine at the Massachusetts General Hospital, and training in Medical Oncology at the Dana Farber Cancer Institute in Boston. In 2003, following postdoctoral studies in functional genomics at the Whitehead Institute in Boston, she returned to Seattle and established her own laboratory at the Fred Hutchinson Cancer Research Center. She is now an Associate Member and Director of the Early Detection Initiative at the Clinical Research Division at the FHCRC, and an adjunct Associate Professor in the Department of Medicine/Division of Oncology at the University Of Washington School Of Medicine. She runs a mass spectrometry-based laboratory focused on developing technologies for biomarker discovery and verification. Her laboratory has a particular focus on developing multiple reaction monitoring mass spectrometry coupled to stable isotope dilution and immunoaffinity enrichment for precise, multiplex quantitation of proteotypic peptides in complex matrices such as plasma.
Dr. Brigitte Simons is the proteomics application lead at AB SCIEX in Toronto. Prior to working at AB SCIEX, Brigitte received her Ph.D. in Chemical Biology at the University of Ottawa. She then completed two research fellowships at the Centre for Biologics Research at Health Canada and the National Institute of Heart Lung and Blood in Bethesda MD. Brigitte has published most recently in the field of phosphorylation-mediated cell signaling in application to kidney physiology jointly with collaborators from the National Institutes of Health.
